Five crews attended, two from West Ashland, one from Broughton, one from Buckingham, one from Olney along with four officers.
Smoke was issuing from a two-storey property on arrival.
The firefighters used six sets of breathing apparatus, two hose reel jets, one main jet, a thermal imaging camera and a positive pressure ventilation fan.
One woman and one girl were located and brought out of the house by the firefighters, who provided immediate emergency care. Sadly both the woman and the girl died at the scene.
Thames Valley Police, South Central Ambulance Service, and Service and Helicopter Emergency Medical Service crews also attended.
A road closure was put in place.
